缺氧诱导因子-1α(hypoxia-inducible factor-1α,HIF-1α)作为唯一一个特异性低氧状态下发挥活性的转录因子,在肿瘤细胞的侵袭、转移中发挥重要作用[3]。波形蛋白(Vimentin)作为上皮间质转化(epithelial-mesenchymal transition,EMT)的标记分子,同时也是HIF-1α的下游靶基因,其表达可受HIF-1α的调控[4]。在本实验中,我们以小干扰RNA(small interference RNA,siRNA)特异性沉默HIF-1α基因,观察其对缺氧状态下人UMM细胞系(OCM-1)中Vimentin表达的影响,初步探讨HIF-1α对葡萄膜黑色素瘤EMT的调控作用及对其侵袭、转移的影响。
